More recently, the essential nature of selenium has become the center of attention, and this element is now known to be required by laboratory animals, food animals (including fish), and humans. Its role as an integral feature of glutathione peroxidase has been established, and other possible functions are under active investigation. This report reviews current knowledge concerning selenium in nutrition, including chemistry, distribution, metabolism, biochemical functions, deficiency signs, and effects of excess intake. For further background, the reader may wish to refer to the earlier reports of the National Research Council: Selenium in Nutrition (1971), Medical and Biological Effects of Environmental Pollutants: Selenium (1976), and Mineral Tolerance of Domestic Animals (1980). Since the previous M.D. Anderson Solid Tumor Oncology Series publication on pancreatic cancer, there have been major advances in our understanding of molecular events which underlie pancreatic cancer development, both in the sporadic and inherited forms. We have seen the development of the first mouse models that accurately recapitulate features of the human disease. Several landmark clinical trials in both resectable and metastatic pancreatic cancer have been completed, raising new questions about the standard of care in this disease. Finally, the era of targeted biologic therapies has engendered new excitement about the prospects of more rapid progress in understanding and successfully treating this dreadful disease. Therefore, it is an appropriate time to review these important advances and outline areas of controversy and question in pancreatic cancer biology and treatment.
